Changes in the leaf structure of plants grown in different conditions have been reported, such as increase in size and density of stomata and reduction in stomatal control, amount of epicuticular wax, and mesophyll thickness, with a high diversity of intercellular spaces. However, these changes are highly variable depending on the physiological and morphological characteristics of each species. This research was undertaken to study the influence of different concentrations of the MT medium, sucrose, vitamins, activated charcoal and gibberellic acid (GA3) on the culture of immature embryos from the crossing between 'Pêra Rio' sweet orange and 'PONCÃ' mandarin. La deformación del mango o “escoba de bruja” es una enfermedad de importancia económica que se encuentra presente en México y en otros países productores. Como agentes causales se ha propuesto a hongos del genero Fusarium y se ha sugerido que el ácaro Aceria mangiferae es portador de dicho hongo. Identification of zygotic and nucellar seedlings in polyembryonic mango cultivars PAB

The objective of this work was to evaluate the occurrence of polyembryony in the mango cultivars Manila and Ataulfo, and to determine whether seedlings cultured in vitro are zygotic or nucelar. Percentage of polyembryony was calculated and the number of embryos in 100 seeds of each cultivar was recorded. 'Manila' exhibited 97% polyembryony with 3.4 embryos per seed, while 'Ataulfo' had 95% polyembryony with 3.2 embryos per seed. Later, 20 seeds of each cultivar were established in vitro, and it was analyzed those in which all embryos germinated (12 seeds from 'Manila' and 7 from 'Ataulfo'). In vitro culture from mature seeds of Passiflora species Scientia Agricola

The genus Passiflora comprises hundred species, mainly native of the South American tropics and rainforests, which are grouped into 21 subgenera. Some species are widely studied for their economic importance and are chiefly cultivated for production of fruit juice. To obtain a continuous source of material for a screening of secondary metabolites, zygotic embryo culture was attempted for 62 Passiflora species, starting from seeds mainly collected in the wild. Twenty nine of these species produced calli, which had very different growth rates. Plants were successfully regenerated from calli of 13 different species. For 25 of the responsive species this is the first report of in vitro culture.

In Vitro Propagation of Heliconia bihai (L.) L. from Zygotic Embryos Acta Botanica

The internal morphology of embryos from immature and mature fruits of Heliconia bihai (L.) L. cv. Lobster Claw Two was examined. Embryos were inoculated into MS media (full MS and ½ MS) and GA3 (0, 2.5 and 5 mg L-1) with either sucrose or glucose. These plantlets were then replicated and transferred to MS medium (full MS or ½ MS) with 0 or 2.5 mg L-1 BAP and their multiplication was evaluated 30 and 45 days after inoculation. The genetic variability of the multiplied plants was estimated using isoenzyme analyses. The internal morphology of the mature embryos revealed their tissues to be in more advanced stages of differentiation than immature embryos. Polyembryony and identification of Volkamerian lemon zygotic and nucellar seedlings using RAPD PAB

The objectives of this work were to evaluate the frequency of polyembryony, and to identify zygotic and nucellar seedlings of Citrus volkameriana using RAPD. Twenty-five polyembryonic and eight monoembryonic seeds were cultivated in vitrofor six months. DNA from seedlings was extracted and used in combination with five RAPD primers to identify zygotic or nucellar origin of the seedlings. Environmental conditions of the year affected significantly (P<0.05) the morphological characteristics of fruitsand the number ofembryos per seed. Polyembryonic seeds ranged from 30.9%, 44.8% to 54.4% over three years. Morphological characteristic was not correlated with polyembryony. 'Ponkan' mandarin (Citrus reticulata Blanco) immature fruits storage Ciência e Agrotecnologia

The aim of this work was to evaluate the effect of 'Ponkan' mandarin (C. reticulata) x 'Pêra' sweet orange (C. sinensis) immature fruits storage and sucrose concentrations on embryos in vitro culture. Fruits with 3 to 4 cm in diameter were harvested and placed inside black polyethylene bags with lateral openings and stored at 5±1ºC during 135 days. Every 15 days a sample was removed, its embryos were excised and individually inoculated in test tubes containing 15 mL of MS medium (Murashige &amp; Skoog, 1962) with sucrose (0, 1.5, 3, 6, 12, 18 and 24 g L-1) and 0.3 mg L-1 GA3 and 1 g L-1 activated charcoal. ABSTRACT Tropical species may require higher temperatures as well as higher growth regulator concentrations for in vitro development. Since these conditions may affect plant metabolism, the objective of this study was to identify how different temperatures and gibberellin concentrations may affect the in vitro development of strelitzia embryos, analyzing the effect on ROS and cytogenetic stability. Zygotic embryos were cultivated on MS medium supplemented with 5, 10 and 20 µM GA3 under temperatures of 25 °C, 30/25 °C and 30 °C.
